PROTECTIVE EFFECTS METAPROT AND ETOMERZOL IN EXPERIMENTAL MODELS POISONINGS WITH HOUSEHOLD POISONS

Abstract

On model of acute intoxication of karbofos, dichloroethane and etilenglikoly (all 256 ± 8.7 mg/ kg) at rats studied mechanisms of protective action of derivatives of tiobenzimidazole (metaprot and etomerzol) 25 and 50 mg/kg. Both preparations renewed of tolerance of physical activity, normalized activity of ASAT, ALAT, reduced levels of bilirubin, a creatinine and urea nitrogen. Impact on processes of lipid peroxidation was expressed in decrease in concentration low-new of dialdehyde and increase of levels restored of glutathione, indicators of endogenous intoxication were thus eliminated. Anti-hypoxemic, antioxidant, aktoprotektorny, energotropny, reparation effects metaprot and etomerzol are proved.

THE ULCEROGENIC EFFECT OF NON-STEROID ANTI-INFLAMMATORy DRUGS AND HEPATOTOXICITy OF NIMESULIDE IN EXPERIMENT ON RATS

Abstract

In animal experimental model the ulcerogenic effect of non-selective and selective inhibitors of cyclooxigenase has been estimated. The all tested remedies influenced morphology of liver and mucosa of stomach. Meanwhile the reaction of each animal on non-steroidal anti-inflammatory drugs was individual. It has been elucidated that ketorolac significantly down-regulated prostaglandin E2 production in gut mucosa. Nimesulide (selective non-steroidal anti-inflammatory drug) significantly up-regulated the activity of aspartat-aminotransferase in serum of experimental rats. The mechanism of nimesulide hepatotoxicity is being discussed.

GASTROPROTECTIVE PROPERTIES OF METAPROT AND DIOSMIN

Abstract

Intragastric administration of acetylsalicylic acid induces development of NSAID-gastropathy in animals. The number of ulcers and the total area of erosive injury in the mucous membrane are studied. Antioxidants/antihypoxants metaprot in a daily dose of 50 mg/kg and diosmin in a daily dose of 100 mg/kg render gastroprotective action. The gastroprotective properties of both drugs are seems to connected with their reparative and antioxidative activity.

METHODOLOGy OF MEMORy STUDy IN PHARMACOLOGICAL CORRECTION OF ITS DISORDERS IN ALCOHOLICS SUFFERED FROM TRAUMATIC bRAIN INjURy

Abstract

The main goal of the current research was to assess memory impairments via learning curves and its restoration during the process of rehabilitative treatment of alcoholic patients having traumatic brain injury with pharmacological correction. We applied a mathematical model Y = B3*exp(-B2*(X-1))+B4*(1-exp(-B2*(X-1))), where X is the trial number; Y is the number of recalled correct words, B2 is the learning rate, B3 is readiness to learn and B4 is ability to learn. Readiness to learn and ability to learn were significantly below in alcoholic patients in comparison with healthy participants. Treatment with vasorelaxants Cinnarizine and Dibazol, central muscle relaxant Baclofen, nootropics Nootropil and Aethimizol, as well as actoprotector Metaprot (Bemithyl) returned readiness to learn up (shortterm memory) to normal values. However, ability to learn (long-term memory) was restored only with Dibazol. Use of 10 words in a list of Luria's memory test is insufficient for exact assessment of the long-term memory and should be increased up to 16 words.

Abstract

The aim of our research is examine features of duirnal type of arterial pressure and parameters of quality of life of patients with asthma combined with hypertension. There were examined 176 patients. They were divided into 3 groups according the main nosology in clinical picture including the group of patients with combined pathology: asthma and hypertension. The features of dairy rate of arterial pressure were exposed in the group with combined nosologies. It was showed that asthma is associated with essential deterioration in patient’s life quality. It depends on expression of bronchoobstructive syndrome.

THE STUDy OF THE NEUROPSyCHOPHARMACOLOGICAL EFFECTS OF THE NEW DERIVATIVE OF GLUTAMIC ACID, THE RGPU-197 COMPOUND

Abstract

Proceeding from the proposition about the high productivity of the search of neuropsychotropic substances among the derivatives of natural neuromediators, a study of neuropsychopharmacological effects of the derivative of the glutamic acid, RGPU-197 compound was carried out. The study was carried out on the he-rats of the Wistar line using the following tests: “open field”, elevated plus maze, conditioned passive-avoidance response, the test of extrapolational deliverance, the test of hanging mice by tails; and also the methods of statistic analysis: rank univariate analysis of Cruscal-Wallace, the Dunn multiple comparison test /the multiple comparison test of Dunn, the x2 criterion. The RGPU-197 compound significantly (p<0,05, p<0,01) decreases the duration of immobilization of the animals and expands the latent period of its approach in the test of hanging mice by tails — it possesses antidepressant activity; increases the quantity of the visits of the centre of the open field, the quantity of attendings of the open hoses of elevated plus maze and the time of stay in them — it shows anxiolytic action; increases the quantity of crossed squares in the open field and the quantity of crossings between the hoses of elevated plus maze — it possesses activating effect; during the displays of the avoidance reflex it expands the latent period of the first period of entering the dark compartment and the quantity of enterings of it in conditioned passive-avoidance response, reduces the latent period of diving under in the test of extrapolational deliverance — it shows nootropic action. According to the intensity of antidepressant action RGPU-197 doesn’t yield to melipramin, anxiolytic and activating effects significantly (p<0.05) excels phenibutum and doesn’t yield to it by nootropic activity. Thus, the new derivative of glutamate, the RGPU-197 compound, possesses the marked antidepressant, anxiolytic and activating effects, as well as nootropic activity.

EFFECT OF AMINOTHIOL ANTIHyPOXANTS ON INTERHEMISPHERIC ASyMMETRy OF THE bRAIN

Abstract

The effect of aminothiol antihipoxants amthizol and its analogue VM-606 on the resistance of the SHR mice males to an acute hypoxia with hypercapnia under conditions of isolated functioning of one of the hemispheres of the brain was studied. Antihypoxic agent amthizol 25 mg/kg increases life time of naïve mice by 46.2%. The drug acted on the sham-operated mice more slightly, increasing of their life only on 28.1% (p<0.01). Administration of amthizol under conditions of functioning of the right hemisphere significantly enhanced (+64.8%) the life time of mice. No antihypoxic effect was registered after administration of amthizol to mice with active left hemisphere: the result was the same as in mice without amthizol. Therefore, antihypoxic effect of amthizol was due to its action on the right (but not the left) hemisphere of the brain. VM-606 possessed more antihypoxic activity in comparison with amthizol. After unilateral cortical inactivation, VM-606 increased life time of mice both in active right and active left hemispheres, but in more degree in active right hemisphere. Thus, interhemispheric differences in resistance of mice to hypoxia with hypercapnia were diminished. Therefore, the differences between amthizol and VM-606 are the followings: amthizol inverts interhemispheric relations in hypoxia whereas VM-606 diminishes them.
