Аннотация

Представлены данные литературы о селективных эстроген-рецепторных модуляторах (СЭРМ). Рассмотрен механизм их действия при связывании с эстрогеновым рецептором, два основных эффекта — эстроген-агонистический и эстроген-антагонистический, реализация которых зависит от экспрессии кофакторов. Подробно описаны наиболее широко применяемые в клинической практике СЭРМ, такие как тамосифен, ралоксифен, торемифен, лазофоксифен и фульвестрант. Показаны их лечебные эффекты, побочные действия, преимущества и недостатки в сравнительном аспекте. Делается заключение, что «идеальный» СЭРМ должен быть эффективным в лечении постменопаузального остеопороза, обладать защитным влиянием на ткань молочной железы, оказывать благоприятные эффекты на сердечно-сосудистую и центральную нервную системы, успешно лечить климактерический синдром с минимумом побочных эффектов.
