Prenatal exposure of N-cholinolytic gangleron compared with M-cholinolytic methamyzil to pregnant rat females at 9-19 days of pregnancy produced the long-term behavioral changes in pubescent rat progenies. Pubescent rat progenies had got the low dynamics of gaining sexual experience and decreased sexual activity with equal disturbance of motivation and coitus. The quantity of males with reduced sexual activity was more compared with the control group. It is suggested that sexual dysfunction of adulthood offsprings is provoked by introduction of gangleron (N-cholinolytic) which was injected at 9-11 and 12-14 days of gestation, and methamyzil (M-cholinolytic) injected at 9-11 days of gestation. We suppose that the regulation of neuronal mechanisms for sexual function is disturbed as a consequence of long-term changings in neurotransmitter activity (first of all in the dopaminergic activity) in the hippocampus and hypothalamus. Therefore, in brain limbic structures was mostly affected. The second reason of the reduced sexual activity in the offsprings is the decrease of the blood testosterone. Сitation: Psychopharmacol Biol Narcol. 2009; 9(1-2): 2530-2539</p></abstract><trans-abstract xml:lang="ru"><p>ЦЕЛЬЮ ДАННОЙ РАБОТЫ являлось изучение отдаленных поведенческих последствий пренатального применения центральных М- и Н-холинолитиков метамизила и ганглерона на половую функцию половозрелых потомств в связи с динамикой онтогенетического развития медиаторных систем мозга.
МЕТОДЫ: Работа выполнена на крысах линии Wistar и их потомствах. Параметры половой функции оценивали в стандартном 15-минутном тесте по Agmo (1997). Концентрацию нейромедиатров и их метаболитов в тканях головного мозга определяли методом ВЭЖХ-ЭД в системе Beckman System Gold. Уровень тестостерона в сыворотке крови определяли методом иммуноферментного анализа. РЕЗУЛЬТАТЫ: Установлено, что пренатальное введение Н-холинолитика ганглерона беременным самкам крыс в различные сроки гестации приводит к отдаленным поведенческим половым изменениям у половозрелых потомств: отмечена низкая динамика приобретения полового опыта и крайне низкая половая активность со значительным нарушением мотивационых и эякуляторных компонентов полового поведения. В экспериментальных группах количество самцов со сниженной половой активностью было значимо выше, чем в группах с контрольным потомством. Пренатальная модуляция ганглероном Н-холинергических механизмов мозга значительно снижает деятельность медиаторных систем ДА, НА, 5-ГТ, которые непосредственно вовлечены в регуляцию мотивации и компонентов коитуса у половозрелых потомств. Результаты эндокринных исследования показали значительное снижение продукции тестостерона ЗАКЛЮЧЕНИЕ: Нарушение половой функции взрослых потомств, вызванное пренатальным введением Н-холинолитика ганглерона на 9-11 и 12-14 сутки гестации и меньшей степени М-холинолитика метамизила на 9-11 сутки гестации, обусловлено нарушением центральных механизмов регуляции половой функции вследствие стойких изменений нейромедиаторной активности в гиппокампе и в гипоталамусе, в большей степени дофаминергической системы, а также значительным снижением уровня тестостерона в крови.
