Ischemic stroke is one of the most common causes of mortality and primary disability. The treatment of ischemic stroke, especially its complications, includes neuroprotective agents among which correctors of mitochondrial dysfunction stand out.</p> <p>Aim of the study. To evaluate VDAC1-dependent mitochondrial effects of compounds containing 4-hydroxy-3,5-di-tretbutyl phenyl substituent in experimental brain ischemia.</p> <p>Material and methods. Brain ischemia was modeled in Wistar rats by irreversible thermocoagulation of middle cerebral artery. After ischemia modeling, the tested compounds (7 objects) and reference drug ethylmethylhydroxypyridine succinate were administered orally (once in day) for 72 hours. After that time, the changes in concentration of ATP, apoptosis-inducing factor, caspase-3, mitochondrial hydrogen peroxide and VDAC1 in brain tissue were assessed in animals.</p> <p>Results. In a study, were shown that the administration of studied compounds in animals with focal ischemia increased the ATP concentration while decreasing the content of mitochondrial hydrogen peroxide and the activity of caspase-dependent and caspase-independent apoptosis reactions. There was also found a decrease of VDAC1 concentration in animals treated with the analyzed compounds, which correlated with the changes of ATP concentration (r=0,89714), apoptosis-inducing factor (r=0,92367) and mitochondrial hydrogen peroxide (r=0,87629).</p> <p>Conclusion. It was concluded that mitochondrial effects of compounds containing 4-hydroxy-3,5-ditrebutyl phenyl which manifested as decreased generation of mitochondrial reactive oxygen species, decreased intensity of reactions of internal apoptosis pathway and increased ATP concentration, were connected with the effect of these compounds on VDAC1 channel activity.</p></abstract><trans-abstract xml:lang="ru"><p>Введение. Ишемический инсульт (ИИ) является одной из самых распространенных причин смерти и первичной инвалидности населения. В лечении ИИ, особенно его осложнений, находят место нейропротекторные средства, среди которых выделяются корректоры митохондриальной дисфункции.</p> <p>Цель исследования. Оценить VDAC1-зависимые митохондриальные эффекты соединений, содержащих 4-гидрокси-3,5-ди-третбутил фенильный заместитель, в условиях экспериментальной ишемии головного мозга.</p> <p>Материал и методы. Ишемию головного мозга моделировали у крыс линии Wistar путем необратимой термокоагуляции средней мозговой артерии. Изучаемые соединения (7 объектов) и референс-препарат этилметилгидроксипиридина сукцинат вводили после воспроизведения ишемии перорально, однократно в день, на протяжении 72 ч. По истечении указанного времени у животных оценивали изменение концентрации АТФ, апоптоз-индуцирующего фактора, каспазы-3, митохондриального пероксида водорода и VDAC1 в мозговой ткани.</p> <p>Результаты. В ходе исследования показано, что применение изучаемых соединений у животных с фокальной ишемией способствовало повышению концентрации АТФ, при снижении содержания митохондриального пероксида водорода и активности реакций каспаза-зависимого и каспаза-независимого апоптоза. Также было установлено снижение концентрации VDAC1 у животных, получавших анализируемые соединения, которое коррелировало с изменением концентрации АТФ (r=0,89714), апоптоз-индуцирующего фактора (r=0,92367) и митохондриального пероксида водорода (r=0,87629).</p> <p>Заключение. На основании полученных данных можно предположить, что митохондриальные эффекты соединений, содержащих в структуре 4-гидрокси-3,5-дитретбутил фенил, которые проявляются в виде уменьшения генерации митохондриальных активных форм кислорода, снижении интенсивности реакций внутреннего пути апоптоза и повышении концентрации АТФ, связаны с влиянием данных соединений на активность каналов VDAC1.</p></trans-abstract><kwd-group xml:lang="en"><kwd>ischemic stroke</kwd><kwd>neuroprotection</kwd><kwd>mitochondrial dysfunction</kwd><kwd>VDAC1</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>инсульт</kwd><kwd>нейропротекция</kwd><kwd>митохондриальная дисфункций</kwd><kwd>VDAC1</kwd></kwd-group><funding-group/></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><mixed-citation>Mendelson S.J., Prabhakaran S.
