Studying the content of these proteins in various parts of the brain of transgenic mice that express increased concentrations of exo- and endogenous HSP will make it possible to clarify the knowledge about the content of these proteins and their distribution in the structures of the central nervous system.</p> <p><bold>The aim. </bold>To develop a method for identifying and analyzing the content of heat shock proteins weighing 70 kDa in sections of brain tissue of transgenic mice using the method of Raman spectroscopy.</p> <p><bold>Material and methods.</bold> To conduct the experiment, were selected 9 mice of the C57BL/6 line (n=3 per group), among them the first group consisted of wild-type mice, the second and third were transgenic mice of the line with an integrated genetic cassette containing copies of the human heat shock protein expressed in the cytosol and into the interstitial space. In this study, Raman spectra of brain sections were measured using a Renishaw inVia Qontor confocal Raman microscope (Renishaw, UK). Raman spectra were recorded from the primary motor cortex and striatum of the brain with an accumulation time of 5 s, the laser excitation wavelength was 633 nm.</p> <p><bold>Results. </bold>The obtained data underwent mathematical preprocessing, which included normalization, removal of outlines and baseline, dimensionality reduction, and noise filtering. There were trained 4 models for classifying Raman spectra, 2 of which showed high accuracy in differentiating spectra from brain sections of mice of the intact group and mice with overexpression of the 70 kDa heat shock protein.</p> <p><bold>Conclusion. </bold>The obtained results demonstrate the feasibility of an attitude to data processing and interpretation, and also allow us to judge the prospects of using the method of Raman spectroscopy to analyze the distribution of heat shock proteins in brain tissue.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> Белки теплового шока (heat shock proteins, HSP) представляют собой группу соединений, которые могут быть использованы в различных отраслях клинической медицины, таких как неврология, фармакология и онкология. Изучение содержания таких белков в различных участках мозга трансгенных мышей, которые экспрессируют повышенные концентрации экзо- и эндогенного HSP, позволит уточнить данные о содержании этих белков и их распределении в структурах ЦНС.</p> <p><bold>Цель</bold> исследования. Разработать способ определения и анализа содержания белков теплового шока массой 70 кДа в срезах ткани головного мозга трансгенных мышей методом классификации спектров комбинационного рассеяния.</p> <p><bold>Материал и методы. </bold>Для проведения эксперимента были отобраны 9 мышей линии C57BL/6 (n=3 в группе), выделенные в три группы: первую группу составили мыши дикого типа, вторую и третью составляли трансгенные мыши линии c встроенной генетической кассетой, содержащей копии человеческого белка теплового шока, секретируемого в цитозоль и в интерстициальное пространство. В данной работе на конфокальном рамановском микроскопе было произведено измерение спектров комбинационного рассеяния срезов головного мозга. Спектры комбинационного рассеяния света регистрировали из области первичной моторной коры и стриатума головного мозга со временем накопления в 5 с, длина волны возбуждения лазера – 633 нм.</p> <p><bold>Результаты. </bold>Полученные данные проходили математическую предобработку, включавшую нормализацию, удаление выбросов и базовой линии, снижение размерности и фильтрацию шума.
