Cockcroft-Gault formula for determining the glomerular filtration rate was used to estimate renal function, according to the results of which all patients were divided into 3 groups depending on the stage of GFR. Percutaneous nephrolithotripsy (PNLT) was performed according to the standard technique using laser treatment. The physical and chemical composition of the nodule was determined by polarization microscopy. All patients underwent morphological and morphometric study of nephrobiopsy specimens obtained at creation of puncture passage during PNLT in patients with nephrolithiasis. Immunohistochemical study was performed using profibrotic marker Anti-TGF beta 1 antibody (1:500, Abcam, UK), Anti-MMP-9 antibody (1:500, Abcam, UK), Anti-FGFR-1 1:50, Abcam, UK).</p> <p><bold>Results</bold><bold>.</bold> Analysis of the results of morphological study revealed a correlation only between morphological manifestations of CPN progression and expression of profibrotic marker TGF-β.</p> <p><bold>Conclusion</bold><bold>.</bold> The increase in TGF-β expression is accompanied by more pronounced atrophic changes in the epithelium of proximal renal tubules, which indicates the triggering of tubulointerstitial fibrosis mechanisms and the role of this marker in the progression of chronic kidney disease in patients with urolithiasis.</p></abstract><trans-abstract xml:lang="ru"><p>Цель исследования: провести оценку экспрессии профибротических маркеров TGF-β1, ММР-9 и FGFR в эпителии проксимальных почечных канальцев у пациентов с мочекаменной болезнью в зависимости от состава конкремента, а также от степени снижения скорости клубочковой фильтрации.</p> <p><bold>Материал и методы.</bold> Проведено комплексное обследование 17 пациентов с мочекаменной болезнью, для оценки почечной функции использовали определение скорости клубочковой фильтрации по формуле Кокрофта-Голта, согласно результатам которой все пациенты были разделены на три группы в зависимости от стадии ХБП. Перкутанная нефролитотрипсия (ПНЛТ) выполнялась по стандартной методике при помощи лазерного воздействия. Физико-химический состав конкремента определялся при поляризационной микроскопии. Всем пациентам проведено морфологическое и морфометрическое исследование нефробиоптатов, полученных при создании пункционного хода в ходе выполнения ПНЛТ пациентам с нефролитиазом. Иммуногистохимическое исследование было проведено с использованием антител к профибротическим медиаторам Anti-TGF beta 1 antibody (1:500, Abcam, UK), Аnti-MMP-9 antibody (1:500, Abcam, UK), Anti-FGFR-1 1:50, Abcam, UK).</p> <p><bold>Результаты.</bold> Анализ результатов морфологического исследования выявил корреляционную взаимосвязь только между морфологическими проявлениями прогрессирования ХПН и экспрессией профибротического маркера TGF-β.</p> <p><bold>Заключение.</bold> Возрастание экспрессии TGF-β сопровождается более выраженными атрофическими изменениями эпителия проксимальных почечных канальцев, что свидетельствует о запуске механизмов тубулоинтерстициального фиброза и о роли данного маркера в прогрессировании хронической болезни почек у пациентов с мочекаменной болезнью.</p></trans-abstract><kwd-group xml:lang="en"><kwd>urolithiasis</kwd><kwd>glomerular filtration rate</kwd><kwd>chronic kidney disease</kwd><kwd>transforming growth factor (TGF-β)</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>мочекаменная болезнь</kwd><kwd>скорость клубочковой фильтрации</kwd><kwd>хроническая болезнь почек</kwd><kwd>трансформирующий фактор роста (TGF-β)</kwd></kwd-group><funding-group/></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><mixed-citation>Zdravoohranenie v Rossii. 2021: Stat.sb. - M.: Rosstat.
