Polycomb response elements (PREs) maintain epigenetic memory within regulatory domains and are often located near insulators, enhancing their blocking activity. The <italic>bxd</italic> PRE is a well-characterized silencer that regulates the <italic>bxd/pbx</italic> domain of the <italic>Ubx</italic> gene in the first abdominal segment. We used a boundary replacement strategy to assess the insulator activity of the <italic>bxd</italic> PRE. Substituting the <italic>bxd</italic> PRE for the <italic>Fab-7</italic> boundary, which separates the <italic>Abd-B</italic> regulatory domains in the BX-C, revealed that the <italic>bxd</italic> PRE has weak insulator activity.</p></abstract><trans-abstract xml:lang="ru"><p>Автономное функционирование регуляторных доменов в комплексе Bithorax (BX-C) <italic>Drosophila melanogaster</italic> обеспечивается границами (инсуляторами), которые предотвращают взаимодействия между энхансерами и промоторами. Сайленсеры (Polycomb response elements) поддерживают эпигенетическую память в пределах регуляторных доменов и часто расположены рядом с инсуляторами, усиливая их инсуляторную активность. <italic>bxd</italic> PRE — это хорошо известный сайленсер, который регулирует активность домена <italic>bxd/pbx</italic> гена <italic>Ubx</italic> (один из генов BX-C) в первом абдоминальном сегменте. Для исследования инсуляторной активности <italic>bxd</italic> PRE мы применили стратегию замены границы.
