It has recently been shown that the PCID2 subunit of TREX-2 is responsible for the specific recognition of mRNA by the TREX-2 complex. The majority of the protein contains the PCI domain, which has surfaces for RNA binding. The PCI domain includes the central region of the protein, which has a surface for non-specific RNA binding, M-PCID2, and the C-terminal part of the PCI domain and the C-terminal part of the protein, C-PCID2, which has a surface for specific RNA recognition. The N-terminal part of PCID2 contains a region whose function is unknown. Because the TREX-2 complex binds only a specific mRNA and only at a specific stage, we hypothesized that the N-terminal part of PCID2 might modulate the binding of C-PCID2 to RNA by binding to it and covering its RNA binding domain. We showed that the N-terminal region interacts with C-PCID2.The binding of C-PCID2 to RNA is then not impaired. Also, the binding of C-PCID2 to RNA does not disrupt its interaction with the N-terminal part of the protein (N-PCID2). Thus, C-PCID2 can interact with N-PCID2 and RNA by different surfaces. This intrinsic interaction is probably necessary at one of the stages of activity of the TREX-2 complex.</p></abstract><trans-abstract xml:lang="ru"><p>Комплекс TREX-2 отвечает за экспорт мРНК из ядра в цитоплазму и состоит из четырех белков. Недавно было показано, что за специфичное распознавание мРНК комплексом TREX-2 отвечает его субъединица PCID2. Большую часть белка составляет PCI-домен, который имеет поверхности для связывания РНК. PCI-домен включает центральную область белка, в которой располагается поверхность для неспецифичного связывания РНК, M-PCID2, и C-концевую часть PCI-домена и С-конца белка, C-PCID2, в которой расположена поверхность для специфичного распознавания РНК. В N-концевой части PCID2 находится район, функции которого не известны. Поскольку комплекс TREX-2 связывает только определенную мРНК и только на определенном этапе, мы предположили, что N-концевая часть PCID2 может модулировать связывание C-PCID2 с РНК, связываясь с ним и закрывая его домен связывания с РНК. Мы показали, что N-концевой район взаимодействует с C-PCID2. Связывание C-PCID2 РНК при этом не нарушается. Также связывание C-PCID2 с РНК не нарушает его взаимодействие с N-концевой частью белка (N-PCID2). Таким образом, C-PCID2 может взаимодействовать с N-PCID2 и РНК различными поверхностями.
