Nanoparticles at concentrations of 0.25, 0.5, 2.5, 5, 12.5, 25, 50, 100, and 200 μg/mL were incubated with human blood mononuclear cells for 24, 48, and 72 hours (5% CO<sub>2</sub> at 37°C). We subsequently determined monocyte viability and fullerenol fluorescence intensity in the monocyte gate using flow cytometry. Nanoparticles were adsorbed/internalized by cells proportionally to their concentration and incubation time. Fullerenol С<sub>60</sub>(ОН)<sub>22–24</sub> at 200 μg/mL reduced monocyte viability by an average of 17%, 30%, and 28% after 24, 48, and 72 hours, respectively. Cell survival inversely correlated with nanoparticle uptake intensity. Concentrations ≤100 μg/mL did not exhibit cytotoxicity even after prolonged exposure, supporting its biomedical applicability.</p></abstract><trans-abstract xml:lang="ru"><p>Изучали влияние фуллеренола С<sub>60</sub>(ОН)<sub>22–24</sub> на жизнеспособность моноцитов периферической крови человека, а также его адгезию/интернализацию этими клетками. Наночастицы в концентрациях 0.25, 2.5, 0.5, 5, 12.5, 25, 50, 100 и 200 мкг/мл инкубировали с мононуклеарными клетками крови человека в течение 24, 48 и 72 часов (5% CO<sub>2</sub> и 37ºC). Впоследствии определялись жизнеспособность моноцитов и интенсивность флуоресценции фуллеренола в гейте моноцитов на проточном цитометре. Установлено, что наночастицы адсорбируются/интернализуются клетками пропорционально их концентрации и времени инкубации. Фуллеренол С60(ОН)<sub>22–24</sub> в концентрации 200 мкг/мл снижал жизнеспособность моноцитов в среднем на 17, 30 и 28% через 24, 48 и 72 часа соответственно. Выживаемость клеток обратно коррелировала с интенсивностью поглощения ими наночастиц.
