The formation of food addiction involves effects on the reward system similar to those of narcotic substances. Glutamatergic inputs can modulate the activity of the mesolimbic dopamine system of the brain, thereby affecting the reward system. In this study, we investigated the mechanism of action of an α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor antagonist on the manifestation of compulsive overeating and on stress-activated eating behavior.</p> <p><bold>AIM:</bold> To study the efficacy of the GluA1 AMPA receptor antagonist IEM-1460 in a model of compulsive overeating and to evaluate its antagonistic activity against glutamate AMPA receptors.</p> <p><bold>METHODS:</bold> Experiments were conducted in vivo on Wistar rats and in vitro on isolated <italic>Danio rerio </italic>neurons. IEM-1460 (3 mg/kg, intraperitoneally) was studied in a model of compulsive hyperphagia with restricted access; stress was induced by electrical foot stimulation (foot-shock). The effect of IEM-1460 on ion currents induced by application of kainic acid (an AMPA receptor agonist) was studied on isolated <italic>Danio rerio </italic>neurons using the patch-clamp technique.</p> <p><bold>RESULTS:</bold> In the model of compulsive hyperphagia with restricted access, administration of IEM-1460 reduced food intake to 10.0 ± 0.9 g, which was significantly lower than the values without additional exposure (15.9 ± 0.5 g) and after foot-shock stimulation (18.6 ± 0.9 g). <italic>In vitro</italic>, IEM-1460 had a blocking effect on AMPA glutamate receptors in the patch-clamp assay.</p> <p><bold>CONCLUSION:</bold> These findings highlight the important role of glutamate AMPA receptors in the formation and maintenance of food addiction. The GluA1 AMPA receptor antagonist IEM-1460 shows high efficacy in the model of compulsive hyperphagia in rats after foot-shock stress, which activates eating behavior. Pharmacological agents based on AMPA receptor antagonists may be effective for correcting compulsive hyperphagia induced by psychogenic stress.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Обоснование.</bold> Пищевая зависимость является более чем актуальной проблемой. При формировании пищевой зависимости происходят воздействия на систему вознаграждения аналогичного таковому у наркотических веществ. Известно, что глутаматергические входы способны модулировать активность мезолимбической дофаминовой системы мозга, влияя на систему вознаграждения. В настоящей работе исследован механизм действия антагониста рецептора α-амино-3-гидрокси-5-метил-4-изоксазолпропионата (AMPAR) на проявление компульсивного переедания и на стресс-активированное пищевое поведение.</p> <p><bold>Цель исследования.</bold> Изучение аддиктивного влияния антагониста GluA1 AMPAR ИЭМ-1460 в модели компульсивного переедания; оценить антагонистическую активность ИЭМ-1460 и в отношении глутаматных АМРАR.</p> <p><bold>Методы.</bold> Эксперименты проведены <italic>in vivo</italic> на крысах линии Wistar и <italic>in vitro</italic> на изолированных нейронах <italic>Danio rerio</italic>. Исследуемые вещества ИЭМ-1460 (в дозе 3 мг/кг, внутрибрюшинно) изучали на модели компульсивного переедания с ограниченным доступом. В качестве стрессового воздействия использовали электростимуляцию конечностей крыс, или foot-shock. На изолированных нейронах рыб <italic>Danio rerio </italic>методом пэтч-кламп исследовали влияние ИЭМ-1460 на ионные токи, индуцированные аппликацией агониста АМРАR — каиновой кислоты.</p> <p><bold>Результаты.</bold> На модели компульсивного переедания с ограниченным доступом введение ИЭМ-1460 уменьшало среднее количество употреблённого продукта до 10,0 ± 0,9 г соответственно, что является статистически значимо ниже значений при отсутствии дополнительного воздействия (15,9 ± 0,5 г) и после электростимуляции конечностей foot-shock (FS) (18,6 ± 0,9 г). <italic>In vitro </italic>на методе пэтч-кламп ИЭМ-1460 оказывают выраженное блокирующее действие на глутаматные AMPAR.</p> <p><bold>Вывод.</bold> Данные исследования подчеркивают важную роль глутаматных AMPAR в формировании и поддержании пищевой зависимости. Антагонист GluA1 AMPAR ИЭМ-1460 демонстрирует высокую эффективность в модели компульсивного переедания у крыс после воздействия foot-shock стресс-активирующего пищевое поведение.
