This behavior, which is similar to addiction, is characterized by uncontrolled food consumption. It is believed to be based on two mechanisms: reduced sensitivity to reward and increased negative affective responsiveness.</p> <p><bold>AIM:</bold> To study the efficacy of the orexin-1 receptor antagonist SB-408124 on compulsive overeating in rats that had experienced early life maternal deprivation.</p> <p><bold>METHODS:</bold> Male rats were separated from their mothers for 180 minutes daily from postnatal days 2 to 12. The experiments used male rats aged 90–100 days. To induce compulsive overeating, the animals were given 1-hour access to a high-carbohydrate diet three times a week for 45 days. The high-calorie food was placed within 5 cm reach 15 minutes before feeding with visual contact. Compulsive behavior was assessed using the marble-burying test. The orexin-1 receptor antagonist SB-408124 was administered intranasally for 7 days at a dose of 1 µg/µL (10 µL into each nostril).</p> <p><bold>RESULTS:</bold> Intermittent consumption of additional high-calorie food induced compulsive overeating in rats. Sexually mature animals that had experienced maternal deprivation stress in early life showed increased signs of compulsive overeating compared with controls (<italic>p</italic> &lt; 0.001). In the marble-burying test, the experimental group receiving the high-calorie diet buried more marbles than the control group (<italic>p</italic> &lt; 0.001). Intranasal administration of the orexin-1 receptor antagonist SB-408124 reduced compulsive overeating in the maternal deprivation model under intermittent high-calorie food consumption compared with the control group (<italic>p</italic> &lt; 0.05); the number of marbles buried decreased to control levels. Standard food consumption did not differ from the control group either before or after drug administration.</p> <p><bold>CONCLUSION:</bold> The findings suggest new pharmacological targets for the treatment of eating disorders, particularly compulsive overeating triggered by early-life stressors.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Актуальность.</bold> Высококалорийная пища, ограничения в питании, психический и физический стресс могут провоцировать приступы компульсивного переедания. Это поведение, сходное с зависимостью, характеризуется неконтролируемым потреблением еды. Считается, что в его основе лежат два механизма: сниженная чувствительность к вознаграждению и усиление негативного аффективного реагирования.</p> <p><bold>Цель исследования.</bold> Изучение действия антагониста рецептора орексина-1 SB-408124 на компульсивное переедание у крыс, перенёсших материнскую депривацию в раннем возрасте.</p> <p><bold>Методы.</bold> Крыс самцов со 2-го по 12-й день после рождения отлучали от матери на 180 мин. В опытах использовали самцов в возрасте 90–100 дней. Для выработки компульсивного переедания животные получали 3 раза в неделю доступ на 1 ч к диете с высоким содержанием углеводов в течение 45 дней. При этом высококалорийную пищу за 15 мин до кормления помещали в 5 см досягаемости при визуальном контакте. Компульсивность в поведении оценивали с помощью теста закапывания шариков. Антагонист рецептора орексина-1 SB-408124 вводили интраназально 7 дней в дозе 1 мкг/мкл (по 10 мкл в каждую ноздрю).</p> <p><bold>Результаты.</bold> Прерывистое потребление дополнительной высококалорийной пищи вызывало компульсивное переедание у крыс. Половозрелые животные, пережившие стресс материнской депривации в раннем онтогенезе, проявляли повышение признаков компульсивного переедания относительно контроля (<italic>p</italic> &lt; 0,001). В тесте закапывания шариков опытная группа животных, получающая высококалорийное питание, закапывала большее количество шариков, чем контрольная (<italic>p</italic> &lt; 0,01). Интраназальное введение антагониста рецептора орексина-1 SB-408124 снижало проявления компульсивного переедания у крыс на модели материнской депривации в условиях прерывистого потребления высококалорийной пищи по сравнению с контрольной группой (<italic>p</italic> &lt; 0,05); количество закопанных шариков снижалось до контрольных показателей.
