Современные представления о сигнальных путях и защитных механизмах ферроптоза. Биологическая роль распространения сигналов смерти ферроптозными клетками

Аннотация
Ферроптоз как вид неапоптозной регулируемой гибели клеток возникает и развивается при помощи сложных сигналов и регуляторных механизмов. Активные формы кислорода (АФК), используемые для инициации ферроптоза, поступают из различных источников, включая железоопосредованные реакции Фентона, митохондриальные АФК и связанные с мембраной АФК, управляемые семейством белков NOX. Мембранные фосфолипиды, содержащие полиненасыщенные жирные кислоты, являются основными субстратами перекисного окисления липидов при ферроптозе, которое регулируется многими ферментами, включая ACSL4, LPCAT3, ALOXs или POR. Селективная активация путей аутофагической деградации способствует ферроптозу, тем самым увеличивая накопление железа, вызывающее перекисное окисление липидов. Напротив, транспортная система Xc– и ось глутатион – GPX4 играют центральную роль в ограничении перекисного окисления липидов, хотя другие антиоксиданты (коэнзим Q10, тетрагидробиоптерин) также могут ингибировать ферроптоз. Основной ядерный механизм защиты клеток от ферроптоза — активация NFE2L2-зависимого антиоксидантного процесса путем транскрипционной регуляции экспрессии антиоксидантов или цитопротекторных генов. Кроме того, повреждение мембраны, вызванное ферроптозным стимулом, может быть устранено с помощью механизма разрыва мембраны, зависящего от комплекса ESCRT-III.
