Аннотация
Бисфосфонаты (БФ) - класс лекарственных препаратов, проявляющих высокую ингибирующую активность в отношении остеокластов - клеток, отвечающих за резорбцию костной ткани. Благодаря выраженному эффекту подавления процессов разрушения кости, БФ являются одними из наиболее эффективных препаратов, рекомендованных для лечения остеопороза (ОП) - одного из распространенных заболеваний костной ткани, сопровождающегося переломами костей вследствие снижения минеральной плотности костной ткани (МПКТ) и нарушения ее архитектоники. Однако появляется все больше данных о том, что эффективность применения препаратов данного класса не всегда соответствует ожидаемому эффекту, а возникновение нежелательных реакций организма на их прием у части пациентов предполагает существование индивидуальной чувствительности к БФ. В настоящее время появилось много публикаций в области изучения фармакогенетики различных препаратов, в том числе БФ, однако прогноз эффективности лечения, основанный на данных индивидуального генетического профиля, неоднозначен; зачастую результаты, получаемые авторами, противоречивы. В связи с этим представляется актуальной задачей систематизация имеющихся результатов в этой области исследований. В статье представлено обобщение накопленных к настоящему времени актуальных данных по фармакогенетике БФ и описано современное состояние вопроса.
