Аннотация
В последние десятилетия появились лекарственные средства на основе биомакромолекул. По своему воздействию на организм макромолекулы отличаются от малых молекул. Для цитокинов, включая интерфероны и хемокины, а также ферментов, глюканов, антител установлено наличие множества сайтов связывания на одной и той же макромолекуле с соответствующими комплементарными структурами в биологических системах. Свойства плейотропии и избыточности цитокинов, определяемые природы цитокиновых сетей, обуславливают различные функциональные эффекты, т.е цитокины могут оказывать более одного воздействия на одну и ту же клетку. Та же закономерность установлена при изучении воздействия интерферонов. Существует большое количество данных о том, что присутствие нескольких аллостерических сайтов в белках мотивирует разработку аллостерических лекарств-модуляторов активности белка с потенциально более высокой специфичностью и меньшей токсичностью, чем у традиционных ортостерических соединений. То есть существует потенциальное вероятностное множество механизмов действия любой макромолекулы. Использование макромолекул в качестве лекарственных препаратов может запускать тот или иной конкретный механизм их действия из имеющегося множества потенциально существующих. В силу этого, высокомолекулярные лекарственные средства, по-видимому, должны иметь свои особенности при прохождении через регуляторные органы в отличии от низкомолекулярных фармацевтических субстанций.
