The regulatory role of mitochondrial pora and the possibility of its pharmacological modulation

Pozhilova Y.V., Novikov V.E., Levchenkova O.S.

Abstract

The review is devoted to the role of mitochondrial Ca2+-dependent pore (mPTP) in the regulation of metabolic processes in cells under physiological and pathological conditions. The mechanisms of reperfusion injury in the postischemic period involving mPTP are discussed in the paper. The possibilities of pharmacological regulation of metabolic and functional processes in cells by target action on mPTP work are assessed. This approach allows to regulate key cell functions, stimulating either mechanisms of adaptation and survival in extreme conditions or apoptosis. Pharmacological modulators of the mitochondrial pore as drugs have promising value for treatment of ischemic diseases as well as tumor therapy.

Pharmacological correction of extreme effects of infrasound acoustic vibrations

Torkunova O.V., Shabanov P.D.

Abstract

Review devoted the problem of pharmacological defance from infrasound exposure to the organism. The physiological, biophysical and biochemical bases of infrasound action on the organism both of men and animals are presented in the paper. The big practical material on pharmacological drugs action assessed as sound-protectors was analysed. The drugs list includes psychostimulants, nootropics, antioxidants, antihypoxants, vitamines, analgetics, nonsteroid anti-inftamatory drugs, analeptics, adrenoblockers, smasmolytics etc. All drugs divided into two groups: pathogenic and symptomatic type of action. The analysis of efficacy of pharmacological drugs action on the organism functioning changes due to infrasound exposure showed that the most perspective as sound-protectors were the drugs acting on the main pathogenetic chains.

Influence of hypoxen on the data changing of lipid metabolsim in the experimantal dislipoproteinemia

Okunevich I.V., Khnychenko L.K., Shabanov P.D.

Abstract

The hypolipidemic efficiency of antihypoxant hypoxen in the experimantal conditions of alimantary dislipoproteinemia (DLP) in rats and guinea pigs in comparison with lipid-lowering drugs gemfibrozyl and probucol in doclinic investigation was studied. The alimantary models of DLP were created by feeding of rats and guinea pigs of hypercholesterol diet (rich content of food cholesterol, the plant and animal lipids, vitamin D2) during 30 days. The concentration of lipid oxidative products in serum and indicators of lipid metabolism (content of TCh, TG, HDL-Ch) in blood and liver were analysed. The calculation of cholesterol index of atherogenity was used. It was shown that hypoxen in dose 50 mg/kg per os had been decreased the concentration of lipid oxidative products in serum and revealed the pronouced lipid-lowering effect in blood and liver of aninals. The hypolipidemic efficiency of hypoxen was in the proximity of gemfibrozyl and probucol. The experimantal results have been shown the possibility of practic using of this active antihypoxant. Hypoxen seemingly to be useful as a hypolipidemic drug in the complex therapy of cardiovascular and atherosclerosis associated diseaseis.

Dynamics of N-acetylglucosamine in rats with acute cold injury

Bondarev Y.V., Shtrygol S.Y.

Abstract

The Treatment and prevention of acute cold injury refers to the important problem of modern medicine and pharmacology and requires in-depth study of the mechanisms of adaptation to low temperatures. For effective treatment and prevention and treatment of cold injury is required to search for new frigoprotective drug. In our studies found pronounced effect in frigoprotection effect glucosamine hydrochloride, superior efficacy of acetylsalicylic acid in the treatment of acute cold injury. Research one of the biochemical mechanisms investigated frigoprotection effect of glucosamine hydrochloride, which is to normalize reserves of N-acetylglucosamine in the liver, kidney and serum of animals.

Central mechanisms of conditioned place preference in rats

Roik R.O., Smirnov A.A., Vinogradov P.M., Potapkin A.M., Lebedev A.A.

Abstract

The purpose of the investigation was to clear the significance of dopamine, GABA, opioids and sodium influx ionic currents of the nucleus accumbens neurons for the reinforcing effects of a number of psychotropic drugs (opiates, opioids, psychostimulants) on conditioned place preference (CPP) in rats. The microcannules were implanted into the nucleus accumbens (the extended amygdala system) of the Wistar male rats to inject the drugs studied (1 μg in 1 μl in volume for each injection). The rats were learned CPP of a one of narcogenics during 8 days. Some drugs, lidocain, a blocker of sodium influx ionic currents, antagonists of GABAA receptors bicuculline, D1 dopamine receptors SCH23390, D2 dopamine receptors sulpiride and opioid receptors naloxone, administered intrastructurally into the nucleus accumbens, were used for pharmacological analysis. The majority of the blockers studied decreased or abolished the reinforcing effects of amphetamine. Activation of reinforcement by means of fentanyl was reversed with bicuculline, lidocain and naloxone but did not change with dopamine antagonists (SCH23390 and sulpiride). None of the blockers studied effect on CPP of sodium ethaminal excluding bicuculline which reduced it. At last, the leu-enkephaline effects were reversed with naloxone and SCH23390, but strengthened with bicuculline. Sulpiride and lidocain did not effect on CPP of leu-enkephaline. Therefore, the different mechanisms (GABA-, dopamine- and opioidergic) controlling the positive conditioned reinforcement are collected in the nucleus accumbens.

Influence of substance πQ1983 on evoked potentials of somatosensory cortex during acute hypoxia development

Sosin D.V., Yevseyev A.V., Pravdivtsev V.A., Yevseyeva M.A., Shabanov P.D.

Abstract

The influence of selenium-containing metal-complex substance πQ1983 on somatosensory cortex bioelectrical activity has been studied on cats (n = 22) during development of acute hypoxia. The condition of acute hypoxia was performed by making of loop including an animal, an apparatus for artificial lung ventilation, and a reservoir for respiration with 5 liter volume. Evoked activities of neurons were provided by direct electrical current irritations applied to the radial nerve of right superior extremity. Couple needle-shaped electrodes were used for mean evoked potentials registration. 100 mg/kg of the substance πQ1983 was introduced in the stomach 180 min before experiment. Also on any steps of the experiments ECG was registered. Containing of oxygen and carbon dioxide were determined by method of gas analysis in air enabled for respiration. It have been established that the substance πQ1983 taking internally increases cortex neurons functional activity period more than triple during acute hypoxia, stabilizes myocardial electrical activity, and helps animals to sustain the action of critical oxygen and carbon dioxide concentrations. Finally, the substance πQ1983 may be related to the high effectively antihypoxants because it significantly rises the resistance of animals with high CNS organization level (cats) to the acute hypoxia even after introduction per os.

Changes in neuronal ionic currents after extracellular and intracellular administration of afobazol and bupivacaine

Vislobokov A.I., Melnikov K.N., Shabanov P.D.

Abstract

The action of anxiolytic afobazol (2-mercaptobenzimidazolum derivative) in concentration 1 mM and local anesthetic bupivacaine in concentrations 0,5-1 mM on ionic currents of isolated neurons of Lymnaea stagnalis and Planorbarius corneus was studied. The method of intracellular dialysis was used in the experiment. Afobazol and bupivacaine after intracellular administration was shown not to possess their inhibition effect of ionic currents, typical for extracellular administration of drugs. It is suggested that pharmacological targets or bindind sits for these drugs are located on external surface of neuronal membrane.
