Immunoregulatory properties of the Interferons I type

Abstract

The type I interferons (IFN) have been the first discovered group of cytokines. However, it keeps extending due to discovery of new members. In the present review new data about the recently revealed members of this family (IFN kappa, lamda and others) are summarized. The characteristics of IFN I type receptors and basic signalling pathways started by the binding molecules of IFN I type with receptors are being considered. The analysis focuses on the mechanisms activated by IFN I type in the immune response to viruses and bacteriae. Connection of IFN I type with others cytokines in various immune reactions, the regulation of the activity of immunocompetent cells by the FN I type, and the role of IFN-a/p in the immune surveillance on tumours have been considered.

Cancer and aging

Abstract

The current opinions on mechanisms of age-related increase in malignant tumor incidence in human and animal are reviewed. Aging may increase or decrease the susceptibility of various tissues to initiation of carcinogenesis and usually facilitates promotion and progression of carcinogenesis. Aging may prodispose to cancer by several mechanisms: tissue accumulation of cells in late stages of carcinogenesis; alterations in homeostasis, in particular, alterations in endocrine and immune systems; telomere instability linking aging and increased cancer risk. Some common genetic processes (e. g. telomere dysfunction, changes in p53 and Rb activity and in DNA repair, accumulation of DNA lesions, genomic instability) play a critical role both in carcinogenesis and aging. As it was predicted by multi-stage model of carcinogenesis, the increase in susceptibility to tumor promoters have been observed in elderly humans and animals. In genetically modified mouse model (transgenic, knockout or mutant) characterized by the aging delay the tumorigenesis was usually alleviated in comparison to wild type animals, whereas in the case of models of accelerated aging the acceleration of the tumorigenesis was observed. Strategy for cancer prevention must include not only measures to minimize the exposure to exogenous carcinogenic agents, but also measures to normalize the age-related alterations in internal milieu.

On the ability of high density lipoproteins to retard oxidation of erythrocyte phospholipids and to remove their peroxidation products from circulation

Abstract

A correlation between cholesterol coefficient of atherogenicity (CA) or a level of high density lipoprotein cholesterol (HDL-Ch) and the content of conjugated dienes (CD) in erythrocytes was studied. There was a significant positive correlation between CA and CD (the Pearson r = 0,427; p = 0,003) and a negative correlation between HDL-Ch and CD (the Pearson r = –0,528; p = 0,0001) in the range of patients’ HDL cholesterol 32-66 mg/dl. In experiments on animals the elimination rate of human oxidized HDL from rabbit circulation after their i. v. administration was higher than that of native HDL. The data obtained support our earlier suggestion on the double protective effect of HDL on the cell membrane: inhibition of membrane phospholipid oxidation and removal of products of phospholipid oxidation from circulation in case oxidation does occur.

Comparative characteristic of the reactions of organ venous vessels to antihypothensive and antihyperthensive drugs

Abstract

In acute experiments on the mongrel anesthetized cats using the methods of venous resistography it was shown, that antihypotensive (dophamine, phenylephrine) and antihypertensive (nytrii nitroprussid, bendazol) drugs, also noradrenaline and isoproterenol produced accordingly constrictory and dilatatory reactions of venous vessels of calf muscle, small intestine and large subcutaneous vein of hind limb of a cat (vena saphena magna). Constrictory reactions of calf muscle veins, has been provoking by dophamin and noradrenaline more (in 1,5-2 times) exceeded the value of the reactions venous vessels of small intestine. Constrictory reactions of calf muscle veine to phenylefrine insignificantly exceeded analogical reactions of intestinal veines. Dilatatory reactions of calf muscle veines, provoked by natrii nitroprussid and bendazol, exceeded (in 2 times) the values of analogical reactions of intestinal veines. The values of dilatatory reactions of venous vessels of two organs on isoproterenol were compared on quantity. Constrictory (dophamine, phenylephrine) and dilatatory (natrii nitroprussid, bendazol) reactions of subcutaneous veine exceeded analogical reactions of intestinal veines in 2-3 times. The conclusion of the study was that the reactivity of calf muscle veines, especialy subcutaneous veine, on antihypotensive (dofamine, phenylephrine) and antihyperthensive (natrii nitroprussid, bendazol) drugs exceeded the reactivity of intestinal veines significantly.

Role of serotoninergic system of the brain in emotional behavior formation in postnatal period of rats

Abstract

5.7-Dihydroxytryptamine (5.7-DHTH; 75, 100 or 150 mkg), producing selective degeneration of serotoninergic neurons in the brain, was injected i. p. to newborn rats on 4th, 10th or 17th day of life, respectively. All rats were growing, and several behaviors were investigated in adult animals: open field, rotation behavior, anxiety in elevated plus maze, aggression, conditioned place preference, differentiation of novel and known alleys of Y-maze. Early postnatal administration of 5.7-DHTH decreased slightly explorative activity, did not effect on rotations and stereotypy, increased anxiety in elevated plus maze, reduced aggression and reinforcing properties of amphetamine in place preference test and disturbed differentiation of novel and known alley in Y-maze in adult rats. These signs were typical for males in more degree compared with females. Therefore, the early postnatal period is the most sensitive to neurotoxin 5.7-DHTH action. This phenomenon is connected with critical periods of development of reinforcing systems of the brain in ontogeny.

Role of 5-HT receptors in the cognitive functions of the brain and hormonal status in the estrogenes deficit

Abstract

The aim of the present study was an assessment of 5-HT1A receptors participation in the behavioral and hormonal responses in the adult intact and ovariectomized (OVX) female rats. For this purposethe influence of chronic administration of 5-HT1A receptors agonist - 8-OH-DPAT (0,05 mg/kg, s.c.) and 5-HT1A receptors antagonist - NAN-190 (0,1 mg/kg, i.p.), injected separately or in a combination with 17p-estradiol (0,5 mg on each animal), within 14 days on the behavioral and hormonal status of the adults intact and ovariectomized (OVX) female rats was investigated. The learning was investigated on the model of passive avoidance per­formance, behavior of animals was estimated in the «open field» test and in the elevated plus maze, levels of hormones (FSH, LH, progesterone, estradiol) were determined by immunoenzyme analysis. Thus, the results obtained testify the involving of serotonergic receptors 1A and 1B subtypes in the mechanisms of behavioral processes realization, and also the hormonal control in the OVX female rats.

Hereditary disturbance hearing, associated with A1555G mutation in the gene 12S rRNA in mitochondrial DNA

Abstract

60 patients with different ethiopathogenic non-syndromic sensoryneural hearing loss were screened for A1555G point mutation in 12s ribosomal RNA gene of mitochondrial DNA, a reason for hereditary non-syndromic hearing loss. Mutation were found in two patients (∼3,3%) with different phenotypic hearing loss: aminoglycoside ototoxicity in the adult age and sensoryneural hearing loss without a clear ethiology from childhood. In both cases we noticed nonsyndromic, postlingual, bilateral, symmetricand progressive sensory neural hearing loss of III-IV degree. As well as the presence of hearing impairment in blood relatives. Similar mutation in mitochondrial gene as a cause different phenotypic variants of sensory neural hearing loss was studied among various ethnic groups and was revealed for the first time among population of the europion region of Russia.

Some pathogenetic interconnection of metabolic activity and structure osseous tissue оп diagnosis and treatment of osteoporosis

Abstract

Basing on their personal and literature findings, the authors see the main causes of osteoporosis rate increase in greater longevity of people in modern world on one side, and in extensive implementation of instrumental methods for risk group examination on the other side. The accuracy and reproducibility of the method of dual-energetic X-ray densitometry of osseous tissue are discussed. It is shown that examination results may be influenced by high metabolic activity of osseous tissue as well as by mineral matrix phase composition. To control the treatment effect, it is recommended to evaluate 35 skeletal sites on a repeated schedule. Regarding the problem of osteoporosis therapy, the authors emphasize that medical agents act mainly over the processes of osteoclast-osteoblast remodeling. There are no means selectively influencing the process of osteocyte remodeling, the latter representing a lower level of adaptation mechanisms responsible for osseous structure changes in response to changing mechanical and regulatory-metabolic influences over bone. A new principle of osteoporosis pharmacological therapy is suggested based on creation of medical agents improving regulation of osteocyte calcium pump.

Personal relations of patients with HIV infection in the course of the disease

Abstract

It is known, that in our country there is epidemic of HIV-infection, and AIDS develops at a late stage of the disease. However, any disease is capable to break a habitual contacts of a man with environmental, and reduces an opportunities of a labour activity, and quite often causes of a reorganization of all personal orientation. We investigated a harmonicity of system of a person’s relations on a technique «An unfinished phrases» in a group of 103 HIV-infection (m: f ~ 1:1, age 18-40 years), at various duration of disease and ways of infection, depending on HAART. By us is shown the following. First, HIV-infection is the disadaptating factor and it breaks integrity of all system of a different relations of the man, that results in increase of a psychological pressure and destruction of his social communications. Secondly, during the first time is sharp experiencing of the fact of disease, a patient can rather adapt for the diagnosis; however, the mental disadaptation again accrues at a significant deterioration of state of a patient’s health accompanying a defeat of his nervous system, but already it is caused by a personal changes and a decrease of criticism (owing to development of a psychoorganic syndrome). Thirdly, despite of existence of interrelation of parameters at various stages and at various duration of disease, by an attentive consideration a distinctions come to light, therefore it is necessary to take into account both these of parameter (i. e., describing both stage and duration of illness) in each patient’s case for optimization of a diagnostic, treatment and rehabilitation process.

Intestinal enzymopathies: diagnostics, clinics, treatment

Abstract

The own and literary data on a number of intestinal ensimopathies, participatig in hydrolysis of carbohydrates are submitted: lactase, sacharase, maltase, isomaltase, cellobiase. Materials on their distribution, including initial (congenital) ensimopathies are submitted. Questions of their clinical picture, diagnostics and treatment are considered. The special attention is given a problem of lactase deficiency. Materials of use of fermental replaceable preparations are given at hypolactasia (lactade, lactrase, tilactase, deaereas). It is shown, that because of insufficient acquaintance of doctors to a problem intestinal ensimopathies in some cases to such persons the diagnosis chronic enteritis or colitis, chronic pancreatitis is established, and they are long and unsuccessfully treated in out-patient and stationary conditions though at this pathology one exception of food intolerant products results in full disappearance of clinical symptoms on the part of an alimentary tract.

Diagnostics of diabetic neuropathy at the preclinical stage

Abstract

The objective for present study was to improve electroneuromyographic (ENMG) diagnostics of subclinical diabetic polyneuropathy (DPN), that will allow to treat this complication earlier and, accordingly, to inhibit its progression and to avoid further development of its severe form. Two groups of children with diabetes mellitus (group I with ENMG-detected DPN, group II - without DPN) were compared with healthy children (the control group). It was found that the longer duration of the disease correlated with higher rating of symptoms of DPN. Data on turns/amplitude analysis of interference electromyogram (EMG) in group I correlated with nerve conduction velocity. In group II there was a tendency to decreased amplitude and number of turns of EMG at normal parameters of nerve conduction.

The economy of lung gas exchange iп schoolchildren living in the Еuгореan North

Abstract

In the winter period, the spirographic examination of 594 children at age 7-18 y. o. born in Arkhangelsk was conducted. Lung volumes, capacity and indices of ventilation have been evaluated. Gas composition of breathed out air has been determined. Lung volumes and indices of ventilation were adjusted to the system BTPS, and the value of O2 consumption and CO2 discharge in a minute - to the system STPD. The quantitative evaluation of the received indices was conducted by comparison of their actual values with proper values.

It has been established that in northern children, both absolute and relative values of O2 consumption are reliably increased in comparison with proper values and with indices of children who live in the middle part of Russia. Low concentration of CO2 in breathed out air has been revealed, but actual values of CO2 discharge in a minute were quite high because of concentrated ventilation. At the same time, the economy of one breathing cycle and one cardiac cycle in children living in the North was decreased.

Consequently, the economy of the respiratory system in northern children in the first link of oxygen regimes of organisms (total lung ventilation) is decreased and the intensity is increased.

Blood mononuclear heat shock protein 72 kDa in estimation of human health status

Abstract

We used data of clinical blood analysis, serum cortisol detection and quantity of stress protein with molecular weight 70 kDa (HSP72) in blood mononuclears to estimate practically healthy young men status. All indexes had been used let us to estimate absence or presence organism strength, which expresses in different degrees. Increasing quantities of HSP72 inducible isoform in blood mononuclears are a result of influence on organism different inside or external factors, which may be ignored by the person. In conclusion an objective estimation of human organism status using white blood picture proposed by L. Kh. Garkavi has a good concordance with intracellular HSP72 level, which is a molecular stress marker.
